Project results

During stage I entitled: „Synthesis and characterization of the precursors for the further
preparation of N / MPs” several activities were carried out as follows:
Act 1.1 – Synthesis and characterization of carboxymethyl chitosan (CMCS)
Act 1.2 – Conjugation of CMCS with CGSPGWVRC and indolicidin peptides
Act 1.3 – Synthesis and characterization of fluorescein-labeled CMCS

The obtained materials were characterized from several points of view for the correct correlation
of the properties with the synthesis parameters.

During stage II entitled: „Design and synthesis of peptides-functionalized nano /
microparticles (2PEP-N / MPs)” several activities were carried out as follows:
Act 2.1 – Preparation of nano/microspheres functionalized with peptides
Act 2.2 – Preparation of nano/microcapsules functionalized with peptides
Two types of peptide-functionalized particles, nano/microsphere and nano/microcapsules, were
prepared.

During stage III entitled: „Characterization of nano / microparticles functionalized with
peptides (2PEP-N / MPs)” several activities were carried out as follows:
Act 3.1 – Physico-chemical characterization and correlation of the structure with the properties of
2PEP-N / MPs
Act 3.2 – Dexamethasone loading and in vitro release studies

During stage IV entitled: „Evaluation of the biomaterial properties of the 2PEP-N/MPs” several
activities were carried out as follows:
Act 4.1 Enzymatic biodegradability studies
Act 4.2 In vitro evaluation of 2PEP-N/MPs interaction with blood components
Act 4.3 Protein adsorption studies
Act 4.4 Bioadhesion tests
Act 4.5 Optimization of the preparation process of 2PEP-N/MPs

In stage V entitled: "In vitro evaluation of 2PEP-N/MPs" several activities were carried out as
follows:
Act 5.1 – Antimicrobial activity of 2PEP-N/MPs
Act 5.2 – Synthesis and characterization of fluorescein-labeled 2PEP-N/MPs
Act 5.3 – In vitro evaluation of the cytotoxic impact of 2PEP-N/MPs on both normal lung cells
and RAW 264.7 macrophages
Act 5.4 – Clonogenic assay of the efficacy of 2PEP-N/MPs therapy on the viability and
proliferation of tested cells
Act 5.5 – Estimation of in vitro cellular uptake of 2PEP-N/MPs

In stage VI entitled: “In vitro evaluation of apoptosis and anti-inflammatory potential of
2PEP-N/MPs and preliminary in vivo tests” several activities were carried out as follows:
Act 6.1 – Investigation of the cell apoptosis process in both normal lung cells and RAW 264.7
macrophage cells under the action of 2PEP-N/MPs
Act 6.2 – Evaluation of the anti-inflammatory potential of 2PEP-N/MPs in model cell cultures
Act 6.3 – Selection of optimal 2PEP-N/MPs for in vivo tests
Act 6.4 – In vivo biodistribution assays of fluorescein-labeled 2PEP-N/MPs
